Effect of SB203580 on the production of NO and PGE2 and activation of AP-1. (a) Levels of NO and PGE2 were determined by Griess assay and EIA from culture supernatants of RAW264.7 cells treated with SB203580 (25 μM) and LPS (1 μg/mL) for 24 (NO and PGE2) h. (a) HEK293 cells cotransfected with plasmid constructs NF-κB-Luc or AP-1-Luc (1 μg/mL each) and β-gal (as a transfection control) were treated with SB203580 (25 μM) in the presence or absence of TNF-α (20 ng/mL) for NF-κB activation or PMA (10 ng/mL) for AP-1 activation.
