Anti-Inflammatory Cytokine Interleukin-4 Inhibits Inducible Nitric Oxide Synthase Gene Expression in the Mouse Macrophage Cell Line RAW264.7 through the Repression of Octamer-Dependent Transcription

Figure 5

The OCT site in the Nos2 promoter region is required for IFNγ- and LPS-induced promoter activity. (a) The diagram shows the wild-type (wt) and mutant sequences of the OCT site (mOCT) in the Nos2 minimum promoter region. The numbers above the promoter region refer to the nucleotide positions relative to the transcription start site of the mouse Nos2 gene. (b) RAW264.7 cells were transiently transfected with the NOS-62 luciferase reporter construct or a mutant construct containing a mutated OCT site (mOCT), as described above. The relative luciferase activities are shown as percentages of the activity in cells transfected with the wild-type construct (pNOS-996) and stimulated with IFNγ and LPS. Each column and bar represents the mean ± SEM of three independent experiments. The asterisks denote a statistically significant difference compared to the cultures treated with IL-4 ( , Student’s test).
