Palmitic Acid Induces Production of Proinflammatory Cytokines Interleukin-6, Interleukin-1 , and Tumor Necrosis Factor- via a NF- B-Dependent Mechanism in HaCaT Keratinocytes

PA induces activation of IKKα and degradation of IκBα in HaCaT cells measured by western blotting. Keratinocytes were untreated or treated with PA (75, 100, 125, and 150 μmol/L) for 24 hours under serum-free conditions. The cells were harvested, and nuclear extracts were analyzed with IKKα by western blot immediately after the treatment (a) and 24 hours after the PA depletion (b). The densitometry values are means ± SD of three independent experiments (c). The protein expression of IκBα immediately after the treatment (d) and 24 hours after the PA depletion (e) were also determined by western blot.
