NAG-1 does not inhibit LPS-induced inflammatory cytokine formation in macrophages. (a) RAW 264.7 cells were plated in 60 mm dishes. After 24 hr, cells were treated with NAG-1 protein (R&D) at the concentrations indicated for 5 hrs and then treated with LPS (5 ng/mL) for 20 hrs. RNA was isolated from the collected cells and used for real-time PCR analysis, using primers specific to the genes indicated. The data are expressed as fold increases compared to untreated cells. Error bars represent standard deviation. (b) RAW 264.7 cells were transfected with empty vector (−) or NAG-1 expression vector (+), and then the cells were treated with 1 μg/mL LPS for 6 hours. RNA was extracted and used for real-time PCR analysis, using primers specific to the genes indicated, using mβ-actin as a control gene.
