Generation of IL-8 and IL-9 Producing CD4+ T Cells Is Affected by Th17 Polarizing Conditions and AHR Ligands

Effect of single cytokine application on the T helper cell cytokine production. Human CD4+ T cells were cultured with anti-CD3/anti-CD28 in the presence or absence of the indicated cytokines for 5 days. CD4+ T cells cultured with anti-CD3 and anti-CD28 were used as control (medium). After restimulation with PMA, Ionomycin, and Monensin for five hours and fixation/permeabilization the cells were stained for IL-17A, IL-22, IL-9, IL-8, and IFN-γ and analyzed by flow cytometry.
