Astragaloside IV Inhibits NF-κB Activation and Inflammatory Gene Expression in LPS-Treated Mice

AS-IV inhibits LPS-induced gene and protein expression of myeloperoxidase (MPO) in mouse lung. For panel (a), mice were randomly assigned to receive i.p. injection of LPS and sacrificed after 1, 3, 8, or 24 h. For panels (b) and (c), mice were treated as described in the legend of Figure 2. Total RNA and cytosolic protein were isolated from lung. Lung MPO gene expression was quantified as described in the legend of Figure 2. Lung MPO protein was determined using an ELISA kit as described in detail in Section 2. The mRNA data are shown as fold of control after normalization to the internal control gene GAPDH. Data shown are means ± SEM of 4 to 5 animals per group. compared to animals treated with LPS only.
