Effect of MyD88 on the expression of IL-1β and TNF-α in SFs. SFs were pre-treatment with or without MIP, then stimulated with LPS. Cell lysates were collected (at 12 h) for analyzing the protein expression by Western blot (a), and the relative expression of pro-IL-1β (b) and TNF-α (c) was calculated. Total RNAs were extracted (at 6 h) for detecting mRNAs of IL-1β (d) and TNF-α (e) by real-time quantitative PCR. The cell-free culture medium from each sample was collected at 12 h, and the mature IL-1β (f) and TNF-α (g) proteins secreted into the cell culture media were measured by ELISA. Data shows all the values from independent samples of , versus nonstimulated controls or cells without MIP treatment.
