Effect of TIE on iNOS and COX-2 expression in LPS/IFNγ-stimulated RAW264.7 cells. (a, b) RAW264.7 cells were plated at a density of 1 × 106 cells in 30 mm dish overnight and treated with varying concentrations of TIE with treatment of LPS (100 ng/mL) and IFNγ (10 U/mL) for 4 h. Total RNA was isolated and subjected to qRT-PCR. β-actin mRNA was used as an internal control for standardization. (c) RAW264.7 cells were plated at a density of 1 × 106 cells in 30 mm dish overnight. TIE was added with the treatment of IFNγ (10 U/mL) plus LPS (100 ng/mL) for 6 h. Whole cell lysates were prepared and subjected to Western blotting. β-actin protein was used as an internal control for standardization.
