Bone Components Downregulate Expression of Toll-Like Receptor 4 on the Surface of Human Monocytic U937 Cells: A Cell Model for Postfracture Immune Dysfunction

Alteration of subcellular distribution of TLR4 protein by bone component (BC) exposure in U937 cells. U937 cells were treated with 0.5–50 ng/mL BC for 24 hr. Total protein, cytoplasmic portion, and membranous portion of TLR4 in U937 cells were detected using western blot analysis. The β-actin and Gαs were used as an internal control. The bar diagram represents quantification of TLR4 expression in U937 cells. Data represent the results of 3 independent experiments (mean ± SEM; between groups).
