PGE2 Elevates IL-23 Production in Human Dendritic Cells via a cAMP Dependent Pathway

The effect of db-cAMP and forskolin and rp-cAMP on IL-23 expression of zymosan-activated Mo-DCs. After being incubated in the presence or absence of rp-cAMP (2 μM) for 30 min, Mo-DCs were then treated for 4 h with PGE2 (2 μM) or solvent control DMSO or db-cAMP (50 μM) or forskolin (50 μM). Then the cells were incubated with zymosan (20 μg/mL) for 24 h. Expression levels of IL-23 protein were determined by ELISA 24 h after stimulation. Data represent mean ± SEM of 5 separate experiments. versus zymosan alone, versus zymosan alone, and versus PGE2 plus zymosan treatment.
