Krüppel-Like Factor 4 Is a Regulator of Proinflammatory Signaling in Fibroblast-Like Synoviocytes through Increased IL-6 Expression

Expression of KLF4 and IL-6 in RA FLSs in response to TNF-α. KLF4 is induced by TNF-α in a time- and dose-dependent fashion. RA FLSs were incubated with 20 ng/mL TNF-α for the indicated time periods, and expression of KLF4 was assessed by real-time PCR (a) and western blotting (b). RA FLSs were incubated with TNF-α at different doses, and expression of KLF4 was assessed by real-time PCR after 3 h (c) and western blotting after 12 h (d). The lower panels in (b) and (d) indicate the ratio of KLF4/GAPDH protein. Time course of RA FLS IL-6 expression in response to 20 ng/mL TNF-α was assessed by real-time PCR (e) and ELISA (f). GAPDH as an endogenous control. Data were expressed as means ± SEM (); in comparison to the untreated cells (Ctrl).
