Altered Expression of IFN-λ2 in Allergic Airway Disorders and Identification of Its Cell Origins

Figure 5

Induction of the expression of IFN-λ2 mRNA in A549 cells by tryptase and agonist peptides of PAR-2. Cells were treated with tryptase (2.0 μg/mL) with or without leupeptin (Leup, 3.0 μg/mL), SLIGKV-NH2 (SLIGKV, 100 μM) with or without FSLLRN-NH2, reverse peptide VKGILS-NH2 (VKGILS, 100 μM), tc-LIGRLO-NH2 (tc-LIGRLO, 100 μM) with or without FSLLRN-NH2 (FSLLRN, 400 μM), and reverse peptide tc-OLRGIL-NH2 (tc-OLRGIL, 100 μM). Total cellular RNA was isolated and reversely transcribed to cDNA, and the cDNA was used for quantitative real-time PCR analysis. The data were normalized to the housekeeping gene (β-actin gene) and were expressed as mean ± SE fold of control for four separate experiments performed in duplicate. compared with the baseline control. compared with the response to the corresponding stimulus alone.
