Effect of LPE on phosphorylation or expression of rate-limiting enzymes in the arachidonate cascade. RBL-2H3 cells were seeded on a 6-well plate (5 × 10⁵ cells/well) in MEM-α with 10% FBS at 37°C overnight and further incubated with DNP-IgE for 24 h. IgE-sensitized RBL-2H3 cells were preincubated with LPE (0 to 350 μg/mL) prior to antigen challenge. The above cells were washed with 1x DPBS and lysed with cell lysis buffer. The expression of p-cPLA₂, p-5-LO, COX-2, or β-actin was determined as described in Section 2. Similar results were obtained in three independent experiments. versus DNP-HSA-treated group.
