Experimental Porcine Toxoplasma gondii Infection as a Representative Model for Human Toxoplasmosis

IFN-γ induces IDO activity and antiparasitic effects in porcine PCP-R cells. 3 × 105 PCP-R cells were stimulated with IFN-γ (100 ng/ml). In some groups, the IDO inhibitor 1-methyltryptophan (1-MT) was supplemented (100 μg/ml). Kynurenine concentration was determined in the supernatants using Ehrlich’s reagent after three days of stimulation (a). Identical cell cultures were stimulated and afterwards infected with T. gondii (3 × 104 ME49 tachyzoites per well). In the control group, L-tryptophan (L-Trp) (100 μg/ml) was added at the time of infection. After three days, toxoplasma growth was monitored using 3H-uracil incorporation (b).
