Expression of hBD-3 is induced in primary human keratinocytes by PRGF treatment. (a) Primary keratinocytes were treated with different PRGF concentrations (PRGF 1 : 50, 1 : 20, and 1 : 10 diluted in cell culture medium) for 24 hours. Subsequently, RNA was isolated and reverse-transcribed in cDNA, and hBD-3 gene expression was analyzed by real-time PCR. (b) HBD-3 promoter activation was analyzed in keratinocytes transfected with a hBD-3 promoter luciferase plasmid after stimulation with PRGF (diluted to 1 : 10 in cell culture medium) for 24 hours (, , ns = not significant, Student’s t-test).
