TSG-6 Downregulates IFN-Alpha and TNF-Alpha Expression by Suppressing IRF7 Phosphorylation in Human Plasmacytoid Dendritic Cells

TSG-6 reduced the activation of IRF7 in GEN2.2 cells. GEN2.2 cells were treated with TSG-6 (1000 ng/mL) overnight, followed by stimulation with CpG-A (2 μM) (a) or R837 (10 ng/mL) (b) for 30 min. The cells were then fixed, permeabilized, and stained to detect phosphorylated IRF7 (phosphor-IRF7) at Ser477 and Ser479 by flow cytometry.
