IL-17 Receptor Signaling Negatively Regulates the Development of Tubulointerstitial Fibrosis in the Kidney

MMP2 and tPA expressions are compromised in the obstructed kidney of Il17ra−/− mice. WT and Il17ra−/− mice (–11) were subjected to UUO. At day 7 p.i., obstructed and non-UUO kidneys were evaluated for mRNA expression of (a) Il6, Tnfα, Cxcl1, Cxcl2, and Cxcl5, (b) Tgfβ, (c) Mmp9, (d) Mmp2, and (e) tPA by qPCR. (f) Total kidney extracts were subjected to gelatin zymography to assess for MMP9 and MMP2 activities at day 7 post-UUO. The lanes were run on the same gel but were noncontiguous (white line).
