Inhibitory effect of APO on ASC phosphorylation. J774A.1 cells or BMDMs were stimulated with LPS (37 ng/mL) for 8 h and treated with sodium orthovanadate (1 mM) for 3 h. The ASC was enriched by immunoprecipitation. (a) Immunoblot analysis of phosphor-ASC in immunoprecipitates using antibody to phosphorylated tyrosine. ASC was blotted to validate the efficiency of the immunoprecipitation. (b-c) Immunoblot analysis of phospho-ASC in whole cell lysates of (b) J774A.1 or (c) BMDMs using antibody against phospho-specific ASC.
