miR-23b Negatively Regulates Sepsis-Induced Inflammatory Responses by Targeting ADAM10 in Human THP-1 Monocytes

miR-23b alleviates LPS-stimulated ADAM10 expression, inflammatory cytokines production, and apoptosis in THP-1 cells. THP-1 cells were infected by LV3-miR-23b mimics to overexpress miR-23b and then were stimulated by LPS (500 ng/mL) for 12 h. The expression of miR-23b and ADAM10 protein in THP-1 cells was detected by qRT-PCR and western blotting, respectively (a, b). The supernatant concentrations of inflammatory cytokines (IL-1β, IL-6, and TNF-α) from THP-1 cells were measured by ELISA assay (c–e). Apoptotic cell rates were detected by using flow cytometric analysis of Annexin V-FITC/PI staining (f, g). Each experiment was independently repeated at least three times. , , and .
