Saponin from Periploca forrestii Schltr Mitigates Oxazolone-Induced Atopic Dermatitis via Modulating Macrophage Activation

Figure 7

Immunomodulatory effects of PFS on LPS-induced M1 macrophage polarization. (a) BMDMs in 96-well plates ( cells/well) were cultured in the presence or absence of different concentrations of PFS for 24 h; then, 100 μL of 5 mg/mL MTT was added and incubated for 4 h, the formazan formed in the intact cells was dissolved with 150 μL/well dimethyl sulfoxide (DMSO) at 37°C for 10 min, the OD was measured at 490 nm, and cell viability was calculated (). (b) SDS-PAGE protein bands and (c) the optical densities; BMDMs in 6-well plates ( cells/well) were pretreated with PFS at 2 μg/mL and 10 μg/mL for 12 h and stimulated with LPS (500 ng/mL) for 0.5 h; then, protein samples were collected. The expression of macrophage-associated factors was detected using western blot (). The results were , and versus the vehicle group. (d) Macrophage polarization and nuclear localization of p65 were performed by immunofluorescence staining. Scale bar, 20 μm.
