CypB-CD147 Signaling Is Involved in Crosstalk between Cartilage and FLS in Collagen-Induced Arthritis
Proproliferation and antiapoptotic effects of CIA cartilage explants on CIA FLS. (a) Schematic of coculture experimental study design. CIA cartilage and CIA FLS were cocultured in 48-well plates with physical contact. Articular cartilage explants were incubated in 10% FBS DMEM for two days to adjust to an in vitro setting and cultured in the presence with vehicle, with 10 ng/mL IL-1α, andwith 10 ng/mL IL-1α+10 μM Dex for 3 days before being placed into monolayer FLS for another 2 days. (b) Proproliferation effect of CIA cartilage explants on CIA synoviocytes. Cocultured FLS were collected for CCK-8 analysis. (c, d) Antiapoptotic properties of CIA cartilage explants on CIA FLS. Cocultured FLS were harvested for flow cytometric analysis of cell apoptosis. Results are representative images of experiments from at least three independent experiments with similar findings.
