USP5 targeted and interacted with TRAF6. (a) After infection with lentivirus containing USP5 expression plasmid for 48 hours, RA-FLS were transfected with NF-κB reporter plasmid and phRL-TK plasmid (internal control), together with TRAF6, TAK1, or IKKβ expression plasmid for 24 hours. The activation of the NF-κB promoter was measured by a dual-luciferase reporter gene assay. (b) Endogenous interaction between USP5 and TRAF6 was detected in RA-FLS after IL-1β (10 ng/ml) for indicated times. (c) Myc-USP5 and Flag-TRAF6 plasmids were transfected into RA-FLS for 48 hours; the interaction between USP5 and TRAF6 was examined. .
