Antituberculosis Drugs (Rifampicin and Isoniazid) Induce Liver Injury by Regulating NLRP3 Inflammasomes

NLRP3 inflammasomes were required for INH- and RIF-induced inflammatory responses in rats. Rats were treated with INH and RIF for 28 consecutive days and then treated with an NLRP3-inflammasome inhibitor (INF39 or CP-456773, respectively). (a–c) ELISA assays were performed to evaluate the effects of NLRP3 inhibitors on INH- and RIF-induced inflammatory factors (IL-33, IL-18, and IL-1β) in the serum of each group of rats. (d) The levels of NLRP3, ASC, and cleaved-caspase 1 protein expression were assessed by western blot assays, and the relative levels of the proteins were analyzed based on the gray value.
