Degradation of aggrecan by ADAMTS-5 in vitro. (a) Whole brain lysate of 6-month-old adult WT or C6ST-1 TG mice was digested with increasing amounts of ADAMTS-5. The concentrations of ADAMTS-5 used for digestion were indicated. Degradation of aggrecan was compared by Western blotting. Two high molecular weight aggrecan bands shown by arrowheads in C6ST-1 TG mice were degraded more efficiently by ADAMTS-5 than those of WT mice. (b) The residual amounts of two high molecular weight bands in WT (filled square) and C6ST-1 TG (open circle) mice were expressed as values relative to the no enzyme control.
