14-3-3 regulates presynaptic short-term plasticity by modulating Ca_V2.2 channel properties. 14-3-3 binding reduces cumulative inactivation of Ca_V2.2 channels and sustains Ca²⁺ influx and neurotransmitter release (a). Inhibition of 14-3-3 accelerates Ca_V2.2 channel inactivation and enhances short-term synaptic depression (b).
