NF- B and Matrix-Dependent Regulation of Osteopontin Promoter Activity in Allylamine-Activated Vascular Smooth Muscle Cells

DNA binding activity in control and allylamine vSMCs to a panel of NFκB binding sequences. Nuclear extracts were subjected to EMSA against NF-κB binding domains from several promoters (Ha-ras, rOPN, and rMMP-9), as well as a consensus oligo (Promega). Gels were dried and exposed to film for 48 hours at −80°C with intensifying screen. Arrows denote major NF-κB binding complexes identified. C: control; a: allylamine. Similar results were seen in multiple experiments.
