Induction of Mitochondrial Changes Associated with Oxidative Stress on Very Long Chain Fatty Acids (C22:0, C24:0, or C26:0)-Treated Human Neuronal Cells (SK-NB-E)

Flow cytometric evaluation of the effects of C22:0, C24:0, and C26:0 on superoxide anion production in whole cells and at the mitochondrial level. SK-NB-E cells were cultured for 48 h in the absence (control) or presence of α-cyclodextrin (1 mg/mL) (vehicle) or a VLCFA (C22:0, C24:0, or C26:0) used at 5, 10, and 20 μM. The production of superoxide anion on whole cells (HE-positive cells) (a) and at the mitochondrial level (MitoSOX-positive cells) (b) was determined after staining with DHE and MitoSOX, respectively. Data shown are mean ± SD from three independent experiments conducted in duplicates. Significance of the difference between vehicle- and fatty acid-treated cells is indicated by *(Mann-Whitney test; * ). No significant difference was observed between control and vehicle-treated cells.
