Regulation of hypoxia-induced factor by intracellular reactive oxygen species. Reactive oxygen species positively regulates HIF through enhanced HIF production via activation of NF-κB and PI3K/AKt/4E-BP1 pathway. Meanwhile, ROS inhibits HIF degradation by depleting cellular ascorbate, a cofactor for PHD activity, thus inhibiting HIFα hydroxylation and VHL binding.
