Mitochondrial DNA Instability in Cells Lacking Aconitase Correlates with Iron Citrate Toxicity

mtDNA is maintained in rtg1Δ aco1, rtg2Δ aco1Δ, and rtg3Δ aco1Δ mutant cells. (a) Cells grown exponentially in YPD medium were stained with DAPI and then examined using fluorescence microscopy. Strains as described in (a) as well as rtg1Δ, rtg2Δ, and rtg3Δ single mutants were crossed to a rho0 tester strain (X rho0) of opposite mating type. Diploids were selected and grown on YPD (Dextrose) and YPEthanol (Ethanol) medium. Pictures of cells were taken after 3 days’ growth at 30°C.
