Effect of ARE decoy ODNs on PTS induced HO-1 expression in PC12 cells. (a) Cellular distribution of ARE decoy ODN-FITC. PC12 cells were treated with FITC-conjugated ARE decoy ODNs for 6 h, 12 h, and 24 h, respectively. Transfection and immunohistochemical determination were operated as described in Section 2. Scale of bar is 50 μm. (b) Effect of ARE decoy ODNs on HO-1 induction by PTS. PC12 cells were directly exposed to 16 μg·mL−1 PTS for 24 h, or following the treatment of ARE decoy or mut ODNs, protein was detected by Western blotting using specific antibodies against rabbit HO-1 and β-actin. Relative protein expression level of HO-1 was normalized to that of untreated cells after correction for the expression level of β-actin. The data were reported as mean ± SD ( ). , significantly different from vehicle group. The blots were representative of three independent experiments.
