Apigenin Attenuates Atherogenesis through Inducing Macrophage Apoptosis via Inhibition of AKT Ser473 Phosphorylation and Downregulation of Plasminogen Activator Inhibitor-2

Figure 6

Overexpression of AKT lowered the apoptosis rate induced by apigenin. (a) After 72 h lentiviral transfection of empty vector, Akt wild type, AktS473A, Akt T308A, or AktS473A-T308A, proteins from MPM were extracted for Western blotting. (b) After lentiviral transfection with empty vector, Akt wild type, or AktS473A, cells were treated with apigenin (50 μM) or DMSO (0.1%) for 48 h after 1 h pretreatment with OxLDL (50 μg/mL) and subjected to flow cytometric analysis of cells double stained with Annexin V/PI. Proteins from MPM were extracted for Western blotting. Akt wild type increased the expression of PAI-2 and inhibited the apoptosis induced by apigenin (, ). Results shown are the representative of three independent experiments.
