Naphthoquinone Derivative PPE8 Induces Endoplasmic Reticulum Stress in p53 Null H1299 Cells

H1299 cells were transfected with control siRNA or IRE1 siRNA for 24 h. (a) The cells were then treated with 5 μM PPE8 for indicated time. The cell lysates were immunoblotted with IRE1, JNK, p-JNK, and β-actin antibodies. (b) The cells were then treated with indicated concentrations of PPE8 for 12 h. The cell viability was measured by MTS assay and expressed relative to viability of the cells without PPE8 treatment (100% control value). Similar results were obtained in three independent experiments. Results were expressed as the mean ± SD. Asterisks indicate the values that are significantly different from corresponding control siRNA ().
