Inhibition of the RhoA GTPase Activity Increases Sensitivity of Melanoma Cells to UV Radiation Effects

Proliferation curves for the MeWo, MeWo-RhoA-N19, and MeWo-RhoA-V14 clones exposed to genotoxic-equivalent doses of UV radiation. The cells were seeded at a density of 35 × 103 cells per 35 mm culture dish at 24 h before treatment and were exposed to 50 KJ/m2 UVA, 80 J/m2 UVB, or 4 J/m2 UVC radiation. The cells were collected every 24 h for five consecutive days and were counted daily in a Fuchs-Rosenthal chamber. The graphs are representative of three independent experiments, and the standard deviation and statistical significance are shown for only the fifth day. Two-way ANOVA was performed to compare the mutant clones with the MeWo clone treated according to the same specified conditions. ; ; ; .
