Protective Effect of Peroxisome Proliferator-Activated Receptor α Activation against Cardiac Ischemia-Reperfusion Injury Is Related to Upregulation of Uncoupling Protein-3

Depletion of UCP3 abrogated cell protective effect of WY-14643. Cells were incubated with medium containing 50 μM of WY-14643 for 24 h before 20 h of hypoxia and 2 h of reoxygenation. (a) MTT assay shows that cell viability was increased with treatment of WY-14643 50 μM while the enhanced viability was attenuated in cells transfected with UCP3 siRNA. versus control . (b) Wortmannin, PD98059, or zinc protoporphyrin-IX (ZnPP IX) decreased protective effect of WY-14643. Mitigated cell survival with treatment of Wortmannin, PD98059, or ZnPP IX was less significant compared with that of cells transfected with UCP3 siRNA. versus control; versus HR (+), WY-14643 (+), and UCP3 siRNA (+) .
