High-dose AB decreases Nox2 expression but does not affect other cellular antioxidant or prooxidant enzyme systems. NG108-15 cells were treated with 50 μM PQ with various doses of AB. AB was added 24 hours prior to PQ; cells were analyzed 48 hours after PQ administration. (a) Western blot analysis of the major neuronal prooxidant enzymes NADPH oxidases 2 and 4 (Nox2 and Nox4) and antioxidant enzymes catalase, manganese superoxide dismutase (MnSOD), and copper/zinc superoxide dismutase (CuZnSOD). (b) In-gel activity assay for catalase and SOD demonstrating no change with any treatment course.
