Salidroside Inhibits HMGB1 Acetylation and Release through Upregulation of SirT1 during Inflammation

Salidroside attenuates the acetylation but not phosphorylation of HMGB1 induced by LPS in RAW264.7 cells. The cells were preincubated with salidroside for 2 h and then stimulated with LPS for 4 h, and whole-cell lysates were immunoprecipitated with an anti-HMGB1 antibody. (a) Phosphorylated HMGB1 was detected by an anti-phosphoserine antibody. (b) The acetylation of HMGB1 was analysed by an anti-acetyl lysine antibody through Western blotting. The histogram showed the relative expression of phosphorylated HMGB1 or acetylated HMGB1 normalized to total HMGB1. The experiments were done in triplicate, and data were shown as mean ± SD. compared with the group stimulated with LPS.
