Effect of methotrexate on wild-type (WT) and UNG-OE L. donovani strains. (a) Total DNA isolated from WT and UNG-OE cells that were treated with 250 μM methotrexate for 7 hours (lane 2) and 24 hours (lane 4). Lanes 1 and 3 correspond to untreated cells processed identically. Isolated DNA samples were analysed on 1% agarose gel stained with ethidium bromide, and the inverted picture was presented. (b) The inhibitory concentrations for MTX were determined for both WT and UNG-OE cells. Untreated parasites were used as control in this experiment. . ns, nonsignificant.
