Characterization of vascular function and protective effects of macitentan cotherapy in ISMN-treated mice. Endothelium-dependent relaxation (ACh) was determined by isometric tension studies in aortic ring segments in order to assess endothelial function (a). Endothelium-independent relaxation (ISMN, GTN) was determined by isometric tension studies in aortic ring segments in order to measure nitrate tolerance and cross-tolerance (b, c). Vasoconstriction was determined by prostaglandin F2α (PGF2α) (d). The data are mean ± SEM from aortic ring segments of 15-21 (a), 12-16 (b), 13-19 (c), and 16-24 (d) mice per group. vs. control; vs. ISMN group.
