Effects of cafestol and the Sirt1 inhibitor sirtinol on cyclic-strain-induced inflammatory molecule protein expression in HUVECs. (a) Effects of cafestol on Sirt1 expression. Cells were pretreated with cafestol for 12 h or sirtinol for 2 h and then exposed to cyclic strain for 24 h. Protein expression was examined using Western blotting. The data are representative of three independent experiments. The bar graph shows the fold increase in protein expression compared with control cells. (b) Effects of the inhibition of Sirt1 by sirtinol on protein expression levels as examined using ELISA.
