Resveratrol-Induced Downregulation of NAF-1 Enhances the Sensitivity of Pancreatic Cancer Cells to Gemcitabine via the ROS/Nrf2 Signaling Pathways

Knockdown of NAF-1 increases the sensitivity of pancreatic cancer cells to gemcitabine. (a, b) After being treated with increasing concentrations of gemcitabine (0, 1, 2, 5, 10, and 20 μM) for 24 h, 48 h, or 72 h, the inhibition ratio of the cancer cell proliferation was detected using an MTT assay. (c–f) The effects of siNAF-1 combined with gemcitabine or siControl on Panc-1 and Mia paca-2 cancer cell apoptosis after a treatment with 2 μM gemcitabine for 48 h; apoptosis in the cancer cells was detected by flow cytometry. (g–j) The effects of siNAF-1 or siControl combined with gemcitabine on the colony-forming ability of the two cell lines were determined using a colony formation assay.
