UVA Irradiation Enhances Brusatol-Mediated Inhibition of Melanoma Growth by Downregulation of the Nrf2-Mediated Antioxidant Response

BR specifically inhibits the Nrf2 signaling pathway. A375 cells were treated with BR for 24 hours. (a) Cell survival was assessed using an MTS assay. (b and c) Cells were treated with various concentrations of BR for 24 hours ((b), right; (c) right) or with 50 nM BR for different time interval as indicated ((b), left; (c) left). Total cell extracts were prepared and subjected to Western blot using antibodies for Nrf2, HO-1, NQO1, GSTP1, IκBα, COX2, NF-κB, and actin.
