Antimycin A-Induced Mitochondrial Damage Causes Human RPE Cell Death despite Activation of Autophagy
Figure 6
Autophagy was active in Aa-treated ARPE-19 cells. Treatment with 25 μM Aa for 24 hours led to a marked increase in the protein levels of p62 and LC3-II (a, b). Autophagic vesicles were visible also in the TEM analysis (c) (white asterisks: autophagic vesicles; L: lysosome; scale bar: 1 μm), and cells transfected with a LC3 Tandem plasmid construct showed clear punctae indicating either autophagophores (yellow) or autophagosomes (red) (d) (scale bar: 20 μm; blue: DAPI). Western blot data is combined from three independent experiments with four parallels per group. Cropped regions of representative blots and representative microscopy images are shown. Quantitative results are presented as . , Mann-Whitney test.
