NOX2-Dependent Reactive Oxygen Species Regulate Formyl-Peptide Receptor 1-Mediated TrkA Transactivation in SH-SY5Y Cells

FPR1-mediated phosphorylation of Y751 residue of TrkA provides a docking site for PI3K/Akt pathway activation. SH-SY5Y cells were growth-arrested for 24 hours and (a) stimulated for 2, 5, or 10 minutes with 0.1 μM N-fMLP or (b) pretreated with PTX or cyclosporin H or (c) preincubated with apocynin or (d) with GW441756, before N-fMLP stimulation. Fifty micrograms of whole lysates was immunoblotted with an anti-phospho Akt(S473) (α-pAkt(S473)) antibody. An anti-tubulin (α-tubulin) antibody was used as a control for protein loading.
