Antioxidant and Anti-Inflammatory Properties of Anthocyanins Extracted from Oryza sativa L. in Primary Dermal Fibroblasts

Figure 8

Effects of ANT on the activation of NF-κB transcription factors in RDFs stimulated with H2O2. RDFs were treated with different doses of ANT for 2 h and then exposed to 0.3 mM H2O2. Ten hours after H2O2 exposure, cytoplasmic and nuclear extracts were subjected to western blot analysis. (a) Nuclear extracts were immunoblotted for NF-κB p50 and NF-κB p65. (b) Data are expressed relative to the control. (c) Cytoplasmic extracts from the same RDF preparations were immunoblotted for NF-κB p50 and NF-κB p65. (d) Data are expressed relative to the control. Histone H3 and β-actin were used as a loading control for the nuclear and cytosol fraction, respectively. Representative images of immunofluorescence staining showing NF-κB p50 (e) and NF-κB p65 (f) localization conjugated to FITC (green). Nuclei were stained with DAPI (blue). Scale bars: 50 μm. Results are expressed as of three independent experiments. , , and .
