Effects of 15s-HpETE on the redox state of PTEN in HeLa and MEF cells. (a) HeLa and MEF cells were treated with different ratios () of 10 μM 15s-HpETE and Lipofectamine 2000 transfection reagent for 5 min. (b) MEFs were incubated with 1 mM H2O2 or the mixture of 10 μM 15s-HpETE and Lipofectamine (1 : 1 ratio) for 5 min. Cellular protein extracts were then alkylated with 10 mM NEM and subjected to nonreducing or reducing SDS-PAGE, followed by Western blot analysis using antibodies to PTEN or actin. “-” indicates Lipofectamine 2000 transfection reagent only; “+” indicates the mixture of 15s-HpETE and Lipofectamine 2000 transfection reagent or H2O2.
