Effects of 15s-HETE and 15s-HpETE on the redox state of PTEN in MEF cells. (a) Chemical structures of 15s-HpETE and 15s-HETE. (b) MEF cells were treated with 10 μM 15s-HETE or the mixture of 10 μM 15s-HETE and Lipofectamine 2000 transfection reagent for the indicated times. (c) MEF cells were treated with the mixture of 10 μM 15s-HETE and Lipofectamine 2000 transfection reagent or the mixture of 10 μM 15s-HpETE and Lipofectamine 2000 transfection reagent for the indicated times. Cellular protein extracts were then alkylated with 10 mM NEM and subjected to nonreducing or reducing SDS-PAGE, followed by Western blot analysis using antibodies to PTEN or actin.
