Effects of 15s-HpETE on the redox state of PTEN in Prx III and Prx III MEF cells. (a) MEF cells were treated with the mixture of 10 μM 15s-HpETE and Lipofectamine 2000 transfection reagent for the indicated times. Cellular protein extracts were alkylated with 10 mM NEM and subjected to nonreducing or reducing SDS-PAGE, followed by Western blot analysis using antibodies to PTEN or actin. (b) The intensity of oxidized PTEN was quantitated using ImageJ software.
