FA-97, a New Synthetic Caffeic Acid Phenethyl Ester Derivative, Protects against Oxidative Stress-Mediated Neuronal Cell Apoptosis and Scopolamine-Induced Cognitive Impairment by Activating Nrf2/HO-1 Signaling

Figure 2

Effect of FA-97 on H2O2-induced apoptosis of SH-SY5Y and PC12 cells. SH-SY5Y and PC12 cells were treated with H2O2 (500 μM) and FA-97 (0, 0.25, 0.5, and 1 μM) for 24 h. (a) After being double stained by Annexin V-FITC and PI, the percentage of apoptotic cells was measured by flow cytometry. (b) Protein levels of Bax, Bcl-2, and β-actin in total protein lysates were analyzed by Western blot using the indicated antibodies. (c) The Bcl-2/Bax ratios were represented by densitometric analysis, and β-actin was used as the loading control. (d) The expressions of Cytochrome c, Caspase-9, and β-actin were measured by Western blot analysis. The relative ratios of Cytochrome c (e) and cleaved Caspase-9 (f) were represented by densitometric analysis. Results are representative of three independent experiments and expressed as . compared with the control group; and compared with the H2O2-stimulated group.
