Lycorine Attenuates Autophagy in Osteoclasts via an Axis of mROS/TRPML1/TFEB to Reduce LPS-Induced Bone Loss

Lycorine decreases LPS-induced autophagy in OCs. BMMs were prepared and incubated with RANKL (40 ng/ml) in the presence of M-CSF (30 ng/ml) for 40 h, washed, and then incubated further for the indicated period with or without lycorine (1.6 μM) in the presence of LPS (50 ng/ml) and M-CSF (30 ng/ml). The levels of LC3II and p62 were analyzed after 48 h of LPS stimulation with or without lycorine. Bafilomycin A1 (25 nM) was added 4 h before harvesting to stimulate the accumulation of LC3II. Quantification of LC3II and p62 normalized to β-actin was plotted. and compared with RANKL-pretreated cells treated with PBS (V). # and ### compared with LPS-treated cells. Similar results were obtained in three independent experiments.
