The Specific Inhibition of SOD1 Selectively Promotes Apoptosis of Cancer Cells via Regulation of the ROS Signaling Network

The specific SOD1 inhibition induces apoptosis and hinders the formation of cancer cell colonies. (a) After incubation for 24 h with LD100 (0, 20, and 50 μM), cytometric assays of apoptosis are assessed on RWPE-1 and DU145 cells by Annexin V and propidium iodide staining. (b) After transfection with validated 35 pmol siRNA of SOD1 for 24 h, cytometric assays of apoptosis are assessed on HeLa, DU145, and RWPE-1 cells by Annexin V and propidium iodide staining. (c) Exposure to 50 μM LD100 for 2 weeks hinders the formation of HeLa and DU145 cell colonies, whereas the formation of RWPE-1 cell colonies is not obviously affected.
